Evidence that the two binding sites for trypsin on chicken ovoinhibitor are not equivalent. Dissociation of the complexes with porcine trypsin.

چکیده

Complexes between chicken ovoinhibitor (01) and porcine trypsin were allowed to dissociate in the presence of the active site titrant, j-nitrophenyl @‘-guanidinobenzoate, which reacts extremely rapidly with free active trypsin (T). The rate of the slower, “postburst” reaction with titrant, which is a measure of the rate of dissociation of the complexes, showed a distinct break not observed previously with bovine trypsin (ZAHNLEY, J. C., AND DAVIS, J. G. (1970) Biochemistry 9, 1428-1433). The 01-trypsin complexes dissociate according to the scheme

Preferred binding of bovine and porcine trypsins at two different sites on chicken ovoinhibitor. Reduced dissociation of mixed trypsin complexes.

Dissociation of mixed trypsin (bovine plus porcine trypsin) complexes with chicken ovoinhibitor was used to investigate the nonequivalence of the two binding sites for trypsin on the inhibitor. Previous work has shown that 1 mol of trypsin dissociates much more rapidly than the 2nd from unmixed trypsin complexes, those containing 2 mol of one kind of trypsin, bovine or porcine, per mol of inhib...

Improved functional properties of the ovoinhibitor by conjugating with galactomannan.

The chicken egg white ovoinhibitor, a multi-type proteinase inhibitor, was conjugated with galactomannan through the Maillard reaction in a controlled dry heating state at 60 degrees C and 65% relative humidity. The formation of an ovoinhibitor-galactomannan conjugate during dry heating was confirmed by SDS-PAGE. The resulting ovoinhibitor-galactomannan conjugate showed almost the same inhibito...

Studies of the comparative properties of proteins and enzymes from closely related species are receiving increasing attention as an approach to understanding the relationships betwcen molecular structures and functions (1). Outstanding examples of these are the studies on the hemoglobins by Pauling et al. (2) and the studies on ribonucleases by Rnfinsen et al. (3). The purpose of this approach ...

Enhancement Effect of Trypsin on Permeation of Clindamycin Phosphate Through Third-degree Burn Eschar

Antimicrobial therapy remains to be the most important method of wound infection treatment. Systemically administered antimicrobials may not achieve therapeutic level in wound. On the other hand, in the absence of surgical debridement (due to any reason), most topically applied antimicrobials cannot penetrate the wound in therapeutic amount due to the presence of eschar. Burn eschar is a protei...

The presence of two Fc receptors on mouse macrophages: evidence from a variant cell line and differential trypsin sensitivity

A stable variant of a clone of the P388D1 macrophage line was isolated using four cycles of treatment with mouse IgG2a-rabbit anti-kappa complexes and rabbit complement. The variant had the same Ka and about the same number of sites per cell for IgG2a as the parent line. However, the variant had 10% as many binding sites for rabbit IgG in soluble antigen-antibody complexes, and the affinity of ...
